Анна Якунина родилась 9 октября 1968 года в Москве в семье студентов ГИТИСа. Мать Ольга Великанова стала режиссёром театра им. Станиславского, отец Александр Якунин играл в театре им. Моссовета. После школы поступила в Вагановское хореографическое училище в Санкт-Петербурге, где проучилась 4 года, но, не окончив его, вернулась в Москву. В Москве окончила Школу-студию при ансамбле народного танца п/р Игоря Моисеева. В 1986 году поступила на режиссёрский факультет ГИТИСа (курс Б. Г. Голубовского, актёрская группа), который окончила в 1990 году. После ГИТИСа играла в театре «Сатирикон». В 2004 году была приглашена Марком Захаровым в Ленком и с тех пор входит в труппу Ленкома.
